A AST catalisa a transferência do grupo amina do aspartato para o cetoglutarato com formação de glutamato e oxalacetato.
O oxalacetato é reduzido a malato por ação da malato desidrogenase (MDH), enquanto que a coenzima NADH é oxidada a NAD+.
A atividade enzimática da AST na amostra é calculada com base na redução da absorbância em 340 nm, quando o NADH se transforma em NAD+.
